Research Project: On-line Estimation of PSD in Polymorphic Crystallization

Project Description:
The area of optimization and control of crystallization process is one of the most important research themes in the pharmaceutical industry. Not only because crystallization is the major industrial process used in separation and purification, but also due the lack of understanding and control of the polymorphic crystallization resulting in product failures. This issue is particularly important in the pharmaceutical industry because the bioactivity and stability changes significantly with the different polymorphic forms of a drug. Therefore it is critical to identify all the possible forms of the active compound and investigate the stability and bioactivity of each form because there is strict FDA (Food & Drug Administration) regulation in assuring the purity of the product.

The aim of this project is to investigate how to convert Chord Length Distribution (CLD) to PSD and to obtain the polymorphic composition in real time. The proposed analytical method will use several in-line analytical tools. Experiments will be conducted in batch crystallizer equipped with the analytic measurement capabilities which consists of: (1) Raman Spectroscopy; (2) Focused Beam Reflectance Measurement (FBRM); (3) Process Video Measurement (PVM); (4) Attenuated Total Reflection Fourier Transform Infrared (ATR FTIR); and (5) Temperature and agitation controlled batch crystallizer.
